PPAR in Endothelial Cells Influences High Fat Diet-Induced Hypertension

چکیده
Methods: To evaluate the vascular role of PPAR we used a conditional null mouse model. Specific disruption of PPAR in ECs was created by crossing Tie2-Cre transgenic (T2T ) and PPAR -floxed (fl/fl) mice to generate PPAR (fl/fl)T2T (PPAR E-null) mice. Conscious 8to 12-week-old congenic PPAR (fl/fl)Cre (wild type) and PPAR E-null mice were examined for changes in systolic blood pressure (BP) and heart rate (HR), untreated, after 2 months of salt-loading (drinking water), and after treatment for 3 months with high fat (HF) diet alone or supplemented during the last 2 weeks with rosiglitazone (3 mg/kg/d).

Gamma Causes Cerebral Vascular Dysfunction in Response to a High-Fat Diet Endothelium-Specific Interference With Peroxisome Proliferator Activated Receptor
The ligand-activated transcription factor peroxisome proliferator activated receptor gamma (PPAR ) is expressed in vascular endothelium where it exerts anti-inflammatory and antioxidant effects. However, its role in regulating vascular function remains undefined. We examined endothelial function in transgenic mice expressing dominant-negative mutants of PPAR under the control of an endothelial-...
متن کاملIgG receptor FcγRIIB plays a key role in obesity-induced hypertension.
There is a well-recognized association between obesity, inflammation, and hypertension. Why obesity causes hypertension is poorly understood. We previously demonstrated using a C-reactive protein (CRP) transgenic mouse that CRP induces hypertension that is related to NO deficiency. Our prior work in cultured endothelial cells identified the Fcγ receptor IIB (FcγRIIB) as the receptor for CRP whe...
متن کاملLectin-like oxidized low-density lipoprotein receptor-1 is required for the adipose tissue expression of proinflammatory cytokines in high-fat diet-induced obese mice.
Lectin-like oxidized low-density lipoprotein (LDL) receptor-1 (LOX-1) is a receptor for oxidized LDL, and is strongly expressed in endothelial cells at an early stage of atherosclerosis. LOX-1 expression in adipocytes is induced by PPARgamma (ligands and appears to be involved in adipocyte cholesterol metabolism. However, the role of adipose tissue LOX-1 in high-fat diet-induced obesity is unkn...
متن کاملThe effect of Nitrochalcone on biochemical indicators and PPAR-α gene expression in nonalcoholic male NMRI mice steatosis model
Introduction: Non-alcoholic fatty liver disease (NAFLD) is associated with oxidative stress, inflammation, and decrease in peroxisome proliferator-activated receptor alpha (PPAR-α) expression. Nitrochalcone is effective ingredient of chalcones with anti-inflammatory, anti-cancer and anti-hyperglycemic properties. This study examined the effect of intraperitoneal (IP) administration of nitrochal...
متن کاملEffect of modulation of PPAR-γ activity on Kupffer cells M1/M2 polarization in the development of non-alcoholic fatty liver disease
Abnormal lipid-mediated hepatic inflammatory-immune dysfunction and chronic low grade inflammation play an important role in the pathogenesis of non-alcoholic fatty liver disease (NAFLD). Macrophage polarization is an important mechanism for the regulation of inflammatory response. Since PPAR-γ has emerged as a master regulator of macrophage polarization, we aimed to investigate the lipid-induc...
